From mendeliome: "Gene-disease association between bi-allelic variants and PCH is well established, limited evidence for mono-allelic variants causing ataxia as per Bryony's review."

OMIM: "Pontocerebellar hypoplasia (PCH) represents a heterogeneous group of disorders characterized by an abnormally small cerebellum and brainstem. Pontocerebellar hypoplasia type 4 (PCH4) is characterized by severe course and early lethality (Budde et al., 2008). PCH type 2 (PCH2) is characterized by progressive microcephaly from birth combined with extrapyramidal dyskinesia and chorea, epilepsy, and normal spinal cord findings (Barth, 1993)."

3 similar conditions, ClinGen has not reviewed these phenotypes and MONDO just has pontocerebellar hypoplasia type 2A MONDO:0010190. Genotype-phenotype correlation: the common/recurrent Ala307Ser variant is associated with a ‘milder’ form of PCH (type 2A) when homozygous, while compound heterozygosity for Ala307Ser and another variant (e.g. nonsense) conferred a more severe phenotype (type 4). PCH type 5 is similar to type 4 (PMID: 20952379). Possibly type 4 and 5 should be lumped together.
